BPC-157
BPC-157 (Body Protection Compound 157, CAS 137525-51-0) is a synthetic pentadecapeptide with sequence GEPPPGKPADDAGLV, composed of 15 amino acids for a molecular mass of 1419.6 Da. It represents a partial fragment of a protective gastric protein isolated by Sikiric and colleagues in 1991 at the University of Zagreb, the full human version of which remains unidentified. BPC-157 stands out for exceptional stability in acidic gastric (pH 1-2) and intestinal media, a property distinguishing it from most bioactive peptides and giving rise to its historical acronym.
Identified molecular mechanisms. Preclinical studies — over 400 publications on PubMed — describe pleiotropic pharmacology involving the NO/eNOS axis (vascular and endothelial protection), VEGF-A regulation (angiogenesis), early Egr-1 induction (early growth response 1, repair transcription factor) and 5-HT2A serotonergic receptor modulation. Reports also document activity on dopaminergic and GABAergic systems and interaction with intestinal tight junctions (occludin, ZO-1).
Research models. The preclinical spectrum covers orthopaedic models (Achilles tendon, collateral ligament, fracture; Staresinic 2003), gastrointestinal (ethanol, cysteamine, stress gastric ulcers), inflammatory (TNBS/DSS colitis, periodontitis), cardiovascular (ischaemia-reperfusion), neurological (ischaemic stroke, encephalopathy, peripheral neuropathies) and musculoskeletal. The main research administration routes are intraperitoneal (IP) or subcutaneous, with doses ranging from 10 μg/kg to 10 mg/kg depending on protocols.
